旨在建立以磷酸为催化剂,从人参须根粉中直接提取制备稀有人参皂苷Rg5的方法。通过单因素试验和响应面分析对其制备工艺进行优化,并利用HPLC法对人参提取物中的人参皂苷Rg5进行定量分析。试验结果表明:加样回收率在98.23%~101.82%,平均回收率为100.67%,RSD为1.21%;最佳提取工艺为以水-95%乙醇=6∶4的混合溶液为提取剂,固液比为1∶40(g/mL),提取液酸浓度0.15 mol/L,提取温度100℃,提取时间6.5 h,人参皂苷Rg5的最高得率为2.17%。
Aimed at the establishment of phosphoric acid as a catalyst from the ginseng fibrous root extract direct preparation of rare ginsenoside Rg5 method. The preparation process was optimized by single factor test and response surface analysis, and the ginsenoside Rg5 in ginseng extract was quantitatively analyzed by HPLC. The results showed that the recoveries were 98.23% ~ 101.82%, the average recoveries were 100.67% and the RSDs were 1.21%. The optimum extraction conditions were as follows: water -95% ethanol = 6: 4 The highest yield of ginsenoside Rg5 was 2.17% when the liquid ratio was 1:40 (g / mL), the acid concentration was 0.15 mol / L, the extraction temperature was 100 ℃ and the extraction time was 6.5 h.
