目的 探讨猴病毒 40 (SV40 )与人脑肿瘤发病的关系。方法 采用聚合酶链反应(PCR)和原位杂交 (ISH)同时检测 30例正常人脑组织和 198例人脑肿瘤组织以及SHG44和BT32 5两株人脑胶质瘤细胞系中SV40DNA序列 ;并对SV40DNA阳性肿瘤组织采用免疫共沉淀和Westernblot检测大T抗原 (Tag)的表达及Tag RB复合物的存在。结果 人脑肿瘤组织PCRSV40DNA阳性率为48.5 % (96 / 198) ,其中胶质瘤 47.2 % (4 2 / 89) ,脑膜瘤 48.8% (2 1/ 43) ,脑垂体腺瘤 5 1.4% (18/ 35 ) ,神经鞘瘤 43.8% (7/ 16 ) ,先天性肿瘤 5 3.3% (8/ 15 ) ,正常人脑组织PCRSV40DNA阳性率为 6 .7% (2 /30 )。SHG44及BT32 5两株细胞中也分别检测出SV40的DNA序列。人脑肿瘤组织SV40DNA阳性率显著高于正常人脑组织 (P <0 .0 1)。经ISH ,仅在 96例PCRSV40DNA阳性的人脑肿瘤组织中检出 87例阳性 ,2例SV40DNA阳性的正常人脑组织中 1例ISH阳性 ,SHG44及BT32 5两株细胞ISHSV40DNA均阳性。SV40DNA定位于肿瘤细胞核 ,阳性细胞呈弥漫或片灶状分布。 96例SV40DNA阳性脑瘤组织Tag表达阳性 75例 ,所有Tag表达阳性瘤组织均发现Tag与RB形成特异性复合物。结论 人脑肿瘤组织中存在SV40感染 ,提示SV40感染与人脑肿瘤有关 ;在人脑肿瘤组织中Tag广泛表达 ,Tag可能是SV
Objective To investigate the relationship between simian virus 40 (SV40) and the pathogenesis of human brain tumors. Methods The SV40 DNA sequences of 30 normal human brain tissues and 198 human brain tumor tissues and two human glioma cell lines SHG44 and BT32 5 were simultaneously detected by polymerase chain reaction (PCR) and in situ hybridization (ISH). Immunoprecipitation and Western blot were used to detect the expression of large T antigen (Tag) and the presence of Tag RB complex in SV40 DNA positive tumor tissues. Results The positive rate of PCRSV40 DNA in human brain tumor tissues was 48.5% (96/198), including 47.2% (42/89) in glioma, 48.8% (21/43) in meningiomas and 51.4% (18) in pituitary adenomas / 35), schwannoma 43.8% (7/16), congenital tumors 53.3% (8/15), normal human brain PCRSV40DNA positive rate was 6.7% (2/30). The DNA sequences of SV40 were also detected in SHG44 and BT32 5 cells respectively. The positive rate of SV40 DNA in human brain tumor was significantly higher than that in normal human (P <0.01). In ISH, 87 positive cases were detected in 96 cases of PCRSV40 DNA positive human brain tumors, 1 case of ISH positive in 2 cases of SV40 DNA positive normal human brain tissues, and ISHSV40 DNA positive in two cases of SHG44 and BT32 5 cells. SV40DNA located in the nucleus of the tumor, positive cells were diffuse or focal distribution. 96 cases of SV40DNA positive brain tumor tissue tagged positive 75 cases, all Tag-positive tumor tissues were found Tag and RB formation of specific complexes. Conclusion There is SV40 infection in human brain tumor tissues, suggesting that SV40 infection is associated with human brain tumors. Tag is widely expressed in human brain tumor tissues.
