目的 从草乌中分离草乌多糖 ,并研究其组成性质。方法 采用DEAE纤维素 (DEAE 5 2 )柱层析和凝胶柱层析分离纯化草乌多糖 ,气相色谱法和薄层层析法测定其多糖组成 ,凝胶过滤法测定其相对分子质量 ,红外光谱测定其苷键类型 ,比色法测定其糖含量。结果 分离得到的草乌多糖 (AconitumkusnezoffiiReichb .,AK)含有鼠李糖、木糖、甘露糖、葡萄糖、半乳糖、阿拉伯糖 ,它们的摩尔比为 1∶1.7∶2 0 3∶1.7∶5 9.7∶3 .5。其糖含量 88%。AK的平均相对分子质量为 2 .8× 10 4。它的红外光谱中有典型的多糖吸收峰 ,并具有α 型糖苷键 ,紫外扫描无核酸和蛋白特征吸收峰。结论 AK首次从草乌块根中提取获得 ,为草乌多糖的主要成分
Objective To separate the polysaccharides from Aconitum kusnezoffii and study its composition. Methods The polysaccharides were separated and purified by DEAE cellulose (DEAE 52) column chromatography and gel column chromatography. The polysaccharide composition was determined by gas chromatography and thin layer chromatography. The relative molecular mass was determined by gel filtration method. Spectrometry was used to determine the type of glycosidic bonds, and the sugar content was determined by colorimetry. The resulting isolated Aconitum Kusnezoffii Reichb. (AK) contained rhamnose, xylose, mannose, glucose, galactose, and arabinose in a molar ratio of 1:1.7:2 0 3:1.7:5 9.7: 3 .5. Its sugar content is 88%. The average relative molecular mass of AK is 2.8 × 10 4 . Its infrared spectrum has typical absorption peaks of polysaccharides, and has α-type glycosidic bonds. UV scanning has no characteristic absorption peaks of nucleic acids and proteins. Conclusion AK was firstly extracted from the roots of Aconitum kusnezoffii and was the main component of Aconitum polysaccharides.
