目的:通过研究去卵巢骨质疏松症(OP)大鼠股骨中Notch1和骨形态发生蛋白9(BMP9)的蛋白表达,探讨摘除卵巢致肾虚OP的病理机制,并研究左归丸的作用机制。方法:摘除雌性大鼠双侧卵巢的方法复制OP模型,采用左归丸对实验大鼠治疗12周,以福善美片作为阳性对照组,正常组、假手术组作为标准对照组,模型组作为空白对照,用ELISA法检测OP大鼠股骨Notch1和BMP9的蛋白表达。结果:正常大鼠股骨中存在Notch1和BMP9的蛋白表达。与正常组比较,模型组大鼠股骨中Notch1和BMP9的蛋白表达水平有明显的下降。左归丸组、福善美组用药12周后,与模型组相比较,左归丸组可显著上调股骨中Notch1和BMP9的蛋白表达水平(P<0.01)。结论:股骨中Notch1和BMP9的蛋白表达下降可能是导致OP发生的机制之一;左归丸能够上调Notch1和BMP9的蛋白表达,表明左归丸对OP有一定的防治作用。
Objective: To investigate the protein expression of Notch1 and bone morphogenetic protein 9 (BMP9) in the femur of ovariectomized rats with osteoporosis (OP) and to explore the pathological mechanism of kidney deficiency induced by ovariectomy and to study the mechanism of Zuo Gui Pill. Methods: The ovariectomized female rats were used to replicate the OP model. The rats were treated with Zuogui Pill for 12 weeks. The FuShanMei tablet was taken as the positive control group, the normal group and the sham operation group as the standard control group. The model group was taken as Blank control was used to detect the protein expression of Notch1 and BMP9 in femur of OP rats by ELISA. Results: The protein expression of Notch1 and BMP9 existed in the normal femur. Compared with the normal group, the protein expression of Notch1 and BMP9 in the femur of the model group decreased significantly. Compared with the model group, Zuoguiwan group could significantly upregulate the protein expression of Notch1 and BMP9 (P <0.01) in the Zuoguiwan group and Fosamax group for 12 weeks. CONCLUSION: The decreased expression of Notch1 and BMP9 in the femur may be one of the mechanisms leading to OP. Zuogui Pill can upregulate the protein expression of Notch1 and BMP9, indicating that Zuogui Pill has some preventive and therapeutic effects on OP.
