目的:观察通脉益智方对血管性痴呆大鼠学习记忆能力和海马突触结构参数变化的影响。方法:采用高脂血症基础上脑缺血再灌注方法建立血管性痴呆动物模型,用Y型电迷宫检测大鼠的学习记忆能力,用透射电镜和形态计量学方法分析药物对血管性痴呆大鼠海马GrayⅠ型突触结构参数变化的影响。结果:通脉益智方对大鼠电迷宫的行为学障碍有明显改善作用,其突触曲率、突触间隙、突触活动区长度、突触致密物质厚度与模型组比较均有显著改善。结论:通脉益智方可减轻高脂血症脑缺血再灌注后大鼠海马CA1区GrayⅠ型突触形态结构的损伤,改善学习记忆能力。
OBJECTIVE: To observe the effects of Tongmai Yizhi Recipe on learning and memory abilities and hippocampal synaptic structural parameters in rats with vascular dementia. METHODS: Animal models of vascular dementia were established by cerebral ischemia reperfusion with hyperlipidemia. The learning and memory abilities of rats were measured by Y-type electric maze. The drugs were analyzed for vascular dementia by transmission electron microscopy and morphometric analysis. The influence of changes of Gray I synapse structural parameters in hippocampus of rats . RESULTS: Tongmai Yizhi Decoction significantly improved the behavioral disorder of rat electric maze, and its synaptic curvature, synaptic cleft, synaptic active zone length, and synaptic dense substance thickness were significantly improved compared with the model group. Conclusion: Tongmai Yizhi Recipe can reduce the damage of Gray I synapse morphological structure in hippocampal CA1 region after hyperlipidemia cerebral ischemia and reperfusion, and improve learning and memory ability.
