目的:建立双山颗粒中绿原酸、芦丁和槲皮苷3种成分的HPLC含量测定方法。方法:色谱柱采用Diamonsil C18(2)柱(250 mm×4.6 mm,5μm);流动相为乙腈-0.2%磷酸溶液,梯度洗脱;检测波长为355 nm;柱温为25℃;流速为1.0 mL.min-1。结果:绿原酸、芦丁和槲皮苷的线性范围分别为0.106~0.636μg(r=0.999 7),0.035 3~0.211μg(r=0.999 4)和0.030 4~0.243μg(r=0.999 5);3成分的平均回收率在99.0%~100.1%。结论:该法简便、准确、重复性好,可用于双山颗粒剂的质量控制。
OBJECTIVE: To establish a HPLC method for the determination of chlorogenic acid, rutin and quercitrin in Shuangshan Granules. METHODS: Diamonsil C18 (2) column (250 mm × 4.6 mm, 5 μm) was used as the mobile phase. The mobile phase consisted of acetonitrile-0.2% phosphoric acid solution with gradient elution. The detection wavelength was set at 355 nm. mL.min-1. Results: The linear ranges of chlorogenic acid, rutin and quercitrin were 0.106 ~ 0.636μg (r = 0.999 7), 0.035 3 ~ 0.211μg (r = 0.999 4) and 0.0304 ~ 0.243μg ); The average recoveries of 3 components ranged from 99.0% to 100.1%. Conclusion: The method is simple, accurate and reproducible. It can be used for the quality control of Shishan granules.
