目的 探讨原癌基因 c- fos在大鼠急性局灶性脑缺血 /再灌注损伤和高压氧 (HBO)治疗后表达的变化。方法 用血管内细丝栓堵大鼠的大脑中动脉 (MCA ) ,制作成局灶性脑缺血动物模型 ,利用免疫组织化学方法 ,观察了 MCA缺血 1 h,再灌注 4、1 1、2 3、71 h c- fos癌基因表达的变化 ,在以上期间同时应用常压 0 .1 MPa纯氧或 0 .2 5 MPa HBO于开始缺血后 2、9、2 1、45和 6 9h分别治疗 1次 (1 h)。结果 缺血后 5 h,c- fos原癌基因在梗塞区皮层、纹状体、视前区均有明显表达 ,持续 1 2~ 2 4h后开始减弱 ,72 h基本消失。 HBO治疗后 ,各时间点 c- fos癌基因在上述区域的表达均明显减弱。结论 HBO可明显抑制大鼠急性局灶性脑缺血 /再灌注损伤区 c- fos癌基因的表达
Objective To investigate the expression of proto-oncogene c-fos after acute focal cerebral ischemia / reperfusion injury and hyperbaric oxygen (HBO) treatment in rats. Methods The middle cerebral artery (MCA) of rat was occluded with intravascular filaments to make a focal cerebral ischemia model. Immunohistochemical method was used to observe the changes of MCA ischemia 1 h, reperfusion 4, 2 3,71 h c-fos oncogene expression changes during the above period while the application of normal pressure 0. 1 MPa pure oxygen or 0.25 MPa HBO at 2,9,2 1,45 and 69h after the onset of ischemia Treatment were 1 (1 h). Results At 5 h after ischemia, c-fos proto-oncogene was significantly expressed in the cortex, striatum and preoptic area of infarcted area. The c-fos proto-oncogene began to weaken after 1-24 h and disappeared after 72 h. HBO treatment, at each time point c-fos oncogene expression in the above regions were significantly reduced. Conclusion HBO can significantly inhibit the expression of c-fos oncogene in acute focal cerebral ischemia / reperfusion injury in rats
