目的 探讨热疗与放疗联合增强顺铂对子宫颈癌细胞的作用与机制。方法 以体外培养的子宫颈癌Hela 细胞株为研究对象,用组内分组设计方法,以恒温水浴热疗、直线加速器放疗,采用原子吸收光谱法检测Hela 细胞内铂(Pt) 含量及DNA- Pt 含量。结果 10 Gy 以下剂量单独放疗未表现出细胞内Pt 量增加,20 Gy 剂量放疗则其量明显增加,提示放疗损伤细胞膜;37 ℃富氧条件下20 Gy 以下剂量放疗未能增加细胞内DNA- Pt 形成,当辅以43 ℃热疗后,不仅细胞内Pt 含量进一步增加,而且细胞内DNA- Pt 形成也增多,提示热疗能促进放疗损伤的DNA 与Pt 结合。结论 热疗在放疗对顺铂化疗增敏过程中发挥了重要的作用。
Objective To investigate the effect and mechanism of hyperthermia and radiotherapy combined with cisplatin on cervical cancer cells. Methods Cervical Hela cell lines were cultured in vitro and treated with constant-temperature water bath hyperthermia and linear accelerator radiotherapy. The intracellular platinum (Pt) content and DNA-Pt content. Results The dose of 10 Gy alone did not show an increase in intracellular Pt content, while the amount of 20 Gy dose radiation significantly increased, suggesting that radiotherapy damage the cell membrane; under 37 Gy oxygen-rich conditions dose radiation below 20 Gy failed to increase intracellular DNA- Pt Formed, when supplemented with hyperthermia at 43 ℃, not only further increase the intracellular Pt content, but also increase the formation of DNA-Pt intracellular, suggesting that hyperthermia can promote radiation-induced DNA damage and Pt binding. Conclusion Hyperthermia plays an important role in the sensitization of cisplatin to radiotherapy.
