目的探讨干扰miR-150表达对鼻咽癌细胞CNE1侵袭的影响。方法使用脂质体将miR-150抑制剂转染CNE1,以无关序列抑制剂作为阴性对照。采用qRT-PCR技术验证miR-150抑制剂转染细胞中miR-150的表达水平;通过MTS法实验观察miR-150表达下调对CNE1细胞侵袭能力的影响。结果转染miR-150抑制剂的CNE1细胞中miR-150的表达量明显下调。转染miR-150抑制剂的CNE1细胞与对照细胞相比,侵袭速度明显减慢,差异有统计学意义(P<0.05)。结论 miR-150能促进鼻咽癌细胞侵袭能力,可能在鼻咽癌的发生和进展中发挥重要作用。
Objective To investigate the effect of interfering miR-150 expression on the invasion of nasopharyngeal carcinoma cells CNE1. Methods Lipofectamine was used to transfect miR-150 inhibitor to CNE1 and irrelevant sequence inhibitors as negative control. The qRT-PCR was used to validate the expression of miR-150 in transfected cells. The MTS assay was used to observe the effect of down-regulation of miR-150 on CNE1 cell invasion. Results The expression of miR-150 in CNE1 cells transfected with miR-150 inhibitor was significantly down-regulated. The invasion rate of CNE1 cells transfected with miR-150 inhibitor was significantly slower than that of control cells (P <0.05). Conclusion miR-150 can promote the invasion of nasopharyngeal carcinoma cells, which may play an important role in the occurrence and progression of nasopharyngeal carcinoma.
