观察了短期注射重组人生长激素(rhGH)对大鼠GH/IGF-Ⅰ轴上相关物质的影响。方法:将32只SD雄性大鼠分为4组:对照组、运动组、GH组和GH+运动组,运动方式为游泳训练。在实验的最后一周给GH组和GH+运动组大鼠注射rhGH,持续7 d,1次/d,注射剂量为100μg/只。检测大鼠血液GH、ghrelin和IGF-Ⅰ浓度。结果:与对照组相比,短期注射rhGH后,大鼠血浆ghrlin浓度明显下降(P<0.01),血清IGF-Ⅰ浓度显著上升(P<0.01),血液IGF-Ⅰ/ghrelin比值明显增大。结论:1)短期注射rhGH使血浆ghrelin浓度降低,提示外源GH对ghrelin有负反馈调节作用;2)验证了前人的发现,使用外源GH可明显提升大鼠血清IGF-Ⅰ水平。
The effect of short-term injection of recombinant human growth hormone (rhGH) on the GH / IGF-I related substances in rats was observed. Methods: 32 SD male rats were divided into 4 groups: control group, exercise group, GH group and GH + exercise group. The exercise mode was swimming training. In the last week of the experiment, GH and GH + exercise rats were injected with rhGH for 7 days and once daily for a dose of 100μg / body. The concentrations of GH, ghrelin and IGF-Ⅰ in blood were detected. Results: Compared with the control group, the concentration of ghlin in plasma decreased significantly (P <0.01) and the serum IGF-Ⅰ concentration increased significantly (P <0.01). The ratio of IGF-Ⅰ / ghrelin in blood increased significantly. Conclusion: 1) Short-term injection of rhGH reduced plasma ghrelin concentration, suggesting that exogenous GH negative feedback regulation of ghrelin; 2) to verify the previous findings, the use of exogenous GH can significantly increase serum IGF-Ⅰ levels in rats.
