目的:观察川芎嗪对紫杉醇所致的神经痛SD大鼠疼痛行为学的影响,并检测大鼠脊髓背角和背根神经节中神经生长因子(nerve growth factor,NGF)的表达。方法:30只成年雄性SD大鼠随机分为4组,各组分别在第1、3、5、7天腹腔注入2.0mg/(kg·d)紫杉醇,A组(n=6)为对照组,第1~14天,B组(n=8)腹腔注入川芎嗪200mg/(kg·d),C组(n=8)口服普瑞巴林3mg/(kg·d),D组(n=8)同时腹腔注射川芎嗪和口服普瑞巴林,剂量同B、C组。分别于用药前1天和用药后第7、12、16、20天测定大鼠的机械缩足反射阈值(MWT),用药后第22天用免疫组化法检测大鼠腰膨大脊髓背角和背根神经节中NGF的表达。结果:相对于A组,B、C、D组大鼠模型分别在第12天、第7天开始出现有统计学意义的MWT升高,并一直稳定持续升高到实验结束,B、C两组疗效相当,相对于C组,D组止痛效果更明显。NGF检测发现对照组表达明显增高,川芎嗪加普瑞巴林减低NGF的表达最明显。结论:川芎嗪能有效防治紫杉醇所致的大鼠神经病理性疼痛,疗效与普瑞巴林相似,与普瑞巴林还有明显的协同作用,作用机理可能是减低中枢神经系统中NGF的表达从而减轻神经损伤,是值得进一步研究的防治化疗神经痛的中药。
Objective: To observe the effect of ligustrazine on the pain behavior of SD rats induced by paclitaxel and to detect the expression of nerve growth factor (NGF) in spinal dorsal horn and dorsal root ganglion. Methods: Thirty adult male Sprague-Dawley rats were randomly divided into 4 groups. Paclitaxel 2.0 mg / (kg · d) was intraperitoneally injected on the 1st, 3rd, 5th and 7th day in each group. CONCLUSION: Tetramethylpyrazine can effectively prevent and treat the neuropathic pain caused by paclitaxel in rats.
