目的探讨感觉神经肽P物质(substance P,SP)对高氧诱导的胎鼠Ⅱ型肺泡上皮细胞(type II alveolar epithelialcells,AECⅡ)转分化的影响。方法剖宫取出孕2l d(足月为22d)SD胎鼠,分离纯化原代培养AECⅡ,采用随机分组法分为:空气暴露组(氧体积分数为0.21)、高氧暴露组(氧体积分数为0.95)、SP干预组,SP干预组于暴露前加入SP1×10-6mol/L,在置于氧体积分数为0.21和0.95中分别暴露12、24、和48h,电镜观察AECⅡ的形态变化;免疫细胞化学染色法和流式细胞仪检测AECII特异性肺泡表而活性蛋白-C(surfactant protein C,SP-C)及I型肺泡上皮细胞(type I alveolar epithelial cells,AECI)特异性水通道蛋5(aquaporin5,AQP5)的表达。结果与空气组比较,高氧暴露后,AECⅡSP-C的表达、SP-C细胞的表达率及荧光指数(flu-orescence index,FI)明显降低,AQP5表达明显增加;而SP干预后,SP-C、AQP5表达都明显增加,形态学的损伤也有明显的改善。结论SP可促进高氧诱导的胎鼠AECⅡ的转分化,在肺损伤修复中可能起重要作用。
Objective To investigate the effect of sensory neuropeptide substance P (SP) on the transdifferentiation of type II alveolar epithelial cells (AECII) induced by hyperoxia. Methods The fetus of fetus with gestational age of 21 days (term of 22 days) were removed from the cesarean section. The primary cultured AECⅡs were isolated and purified. The rats were randomly divided into three groups: air exposure group (oxygen volume fraction 0.21), hyperoxia exposure group Was 0.95). SP intervention group and SP intervention group were added with SP1 × 10-6mol / L before exposure, exposed for 12, 24 and 48h respectively when exposed to oxygen volume fraction of 0.21 and 0.95, respectively. The morphological changes of AECⅡ were observed by electron microscope. Immunocytochemical staining and flow cytometry were used to detect AECII-specific alveolar surfactant protein C (SP-C) and type I alveolar epithelial cells (AECI) 5 (aquaporin5, AQP5) expression. Results Compared with the air group, the expression of AECⅡSP-C, the expression of SP-C and flu-orescence index (FI) were significantly decreased and the expression of AQP5 was significantly increased after hyperoxia exposure. However, SP- C, AQP5 expression were significantly increased, morphological damage also significantly improved. Conclusion SP can promote hyperoxia-induced transdifferentiation of AECⅡ in fetal rats and may play an important role in the repair of lung injury.
