目的:通过补骨脂素对TNFα诱导血管内皮细胞组织因子的影响,以明确补骨脂素在血栓形成过程中的作用,探讨补骨脂素对心血管系统的调节作用。方法:体外培养人脐静脉内皮细胞(human umbilical vein cells,HUVECs),采用MTT方法,检测补骨脂素对HUVECs细胞活力的影响;以肿瘤坏死因子α(tumor necrosis factor-α,TNF-α)刺激HU-VECs,利用ELISA方法检测补骨脂对TNF-α诱导HUVECs组织因子(Tissue Factor,TF)水平的影响。结果:补骨脂素可以增强正常HUVECs活力,最大浓度约为10-7M。在10-7、10-8M浓度下补骨脂素对TNF-α诱导HUVECs分泌TF呈现显著抑制作用(P<0.01和P<0.05)。结论:补骨脂素能够降低TNFα诱导HUVECs的TF释放,降低血栓形成风险。
OBJECTIVE: To investigate the effect of psoralen on the regulation of cardiovascular system through the effect of psoralen on the tissue factor of vascular endothelial cells induced by TNFα in order to clarify the role of psoralen in thrombosis. Methods: Human umbilical vein cells (HUVECs) were cultured in vitro. The effects of psoralen on the viability of HUVECs were detected by MTT assay. Tumor necrosis factor-α (TNF-α) HU-VECs were stimulated to detect the effect of psoralen on the level of Tissue Factor (TF) induced by TNF-α in HUVECs. Results: Psoralen can enhance the activity of normal HUVECs, the maximum concentration is about 10-7M. Psoralen at 10 -7 and 10 -8 M significantly inhibited the secretion of TF of HUVECs induced by TNF-α (P <0.01 and P <0.05). Conclusion: Psoralen can reduce the release of TF from TNFα induced HUVECs and reduce the risk of thrombosis.
