目的 :探讨血管内皮细胞生长因子 (vascularendothelialgrowthfactor,VEGF)在激光诱导兔视网膜下新生血管形成中的作用 ,观察其抗体对新生血管的影响。方法 :高强度氪激光光凝有色家兔视网膜 1、3、7、14d后 ,制备眼球冰冻切片检查眼底病理变化 ,用组织原位杂交法观察VEGFmRNA表达时相和部位的变化。VEGF抗体治疗组 2只眼于光凝后 10min玻璃体内注射 5 μgVEGF抗体 ,14d后行眼底荧光造影并与对侧眼对照观察荧光渗漏变化。结果 :(1)正常对照家兔视网膜中未见VEGFmRNA的表达 ;(2 )激光光凝后 1-3d ,邻近伤口的视网膜节细胞层和内核层VEGFmRNA表达明显增加 :(3 )光凝后 3d ,光凝斑附近的视网膜下有新生血管样结构出现 ;(4 )VEGF抗体治疗组荧光染料渗漏程度明显降低。结论 :VEGFmRNA在激光光凝后的视网膜中表达增多 ,且与光凝斑周围的新生血管化存在时空对应关系 ,提示VEGF在视网膜下新生血管形成过程中具有重要的作用。VEGF抗体对新生血管形成具有明显的抑制作用
Objective: To investigate the role of vascular endothelial growth factor (VEGF) in laser-induced subretinal neovascularization in rabbits and observe the effect of its antibody on neovascularization. Methods: After high intensity krypton laser photocoagulation on the retina of pigmented rabbits for 1, 3, 7, and 14 days, the ocular pathological changes were examined by frozen sections of the eye. The changes of the expression and location of VEGF mRNA were observed by in situ hybridization. VEGF antibody treatment group 2 eyes 10 min after photocoagulation intravitreal injection of 5 μg VEGF antibody, 14 days after fundus fluorescein angiography and contralateral eye control fluorescence leakage changes observed. Results: (1) The expression of VEGFmRNA was not observed in retina of normal control rabbits. (2) The expression of VEGFmRNA in retinal ganglion cell layer and inner nuclear layer adjacent to the wound was increased 1-3 days after laser photocoagulation: (3) , Neovascularization structures appeared under the retina near the photocoagulation spot; (4) Fluorescent dye leakage of VEGF antibody treatment group was significantly reduced. Conclusion: The expression of VEGFmRNA in the retina after laser photocoagulation is increased, and there is a spatiotemporal relationship between the expression of VEGFm and the neovascularization around the photocoagulation spot, suggesting that VEGF plays an important role in the process of subretinal neovascularization.
