目的:研究梓醇(Catalpol)对高糖高脂诱导的人脐静脉血内皮细胞(HUVECs)炎症反应的影响。方法:体外培养人脐静脉血内皮细胞,用CCK-8法检测不同浓度梓醇溶液对HUVECs细胞增殖活性的影响。将细胞分为空白对照组、高糖高脂组(给予33mmol/L葡萄糖+500μmol/L棕榈酸作用48h)、高糖高脂+1 000mg/L梓醇预处理组(梓醇1 000mg/L预处理24h后,给予33mmol/L葡萄糖+500μmol/L棕榈酸作用48h,以下简称梓醇预处理组),收集细胞上清液,用ELISA检测上清液中的炎症因子TNF-α、IL-6的浓度。结果:梓醇作用于HUVECs细胞的最佳浓度为1 000mg/L。高糖高脂组中TNF-α和IL-6浓度较空白对照组均明显增高(t=7.03、6.68,P均<0.01)。与高糖高脂组比较,梓醇预处理组上清液中炎症因子TNF-α及IL-6浓度均下降(t=-2.87、-3.76,P均<0.05)。结论:高糖高脂可诱导血管内皮细胞炎症反应,而梓醇对高糖高脂诱导的炎症反应有一定的改善作用。
Objective: To study the effect of Catalpol on inflammation induced by high glucose and high fat in human umbilical vein endothelial cells (HUVECs). Methods: Human umbilical vein endothelial cells were cultured in vitro. The effects of different concentrations of catalpol on the proliferation of HUVECs were detected by CCK-8 assay. The cells were divided into blank control group, high glucose and high fat group (given 33mmol / L glucose + 500μmol / L palmitic acid for 48h), high glucose + 1000mg / L catalpol pretreatment group (catalpol 1000mg / L The cells were treated with 33mmol / L glucose + 500μmol / L palmitic acid for 48h after pretreatment for 24 hours, then the cell supernatant was collected. The levels of inflammatory cytokines TNF-α and IL- 6 concentration. Results: The optimal concentration of catalpol on HUVECs cells was 1 000 mg / L. The concentrations of TNF-α and IL-6 in high-glucose and high-fat group were significantly higher than those in the blank control group (t = 7.03,6.68, P <0.01). The levels of TNF-α and IL-6 in the supernatant of Catalpol pretreatment group were significantly lower than those in the high-glucose and high-fat group (t = -2.87, -3.76, P <0.05). Conclusion: High glucose and high fat can induce the inflammatory reaction of vascular endothelial cells, while catalpol can improve the inflammatory response induced by high glucose and high fat.
