In several pregnant animals, the corpus luteum produces peptide hormone relaxin and progesterone. Ultrastructural studies reveal that organelles with protein and steroid hormone synthesis in the cytoplasm of luteal cells, where membrane-entrapped particles or vesicles within the cytoplasm of luteal cells, are thought to be the location for the storage of intracellular relaxin. The authors used peroxidase-peroxidase (PAP) immunocytochemistry to study the localization of relaxin in the luteal cytoplasm. The authors used high-purified rat relaxin immunized rabbits, in order to obtain anti-rat relaxin antiserum, and then use a modified PAP immunocytochemistry to observe the luteal cells of pregnant rats
