目的 :探讨HCVNS3丝氨酸蛋白酶与P53蛋白之间的关系 ,为研究NS3丝氨酸蛋白酶在HCV所致肝癌发生发展机制中的作用奠定基础。方法 :构建HCVNS3丝氨酸蛋白酶基因及其N端缺失突变体的重组质粒 ,通过酵母双杂交系统定性及定量检测P53与NS3及共突变体的相互作用。结果 :HCVNS3丝氨酸蛋白酶、N端缺失 15个氨基酸及缺失 30个氨基酸的NS3丝氨酸蛋白酶与P53均有相互作用 ,且相互作用的强度无显著差异。结论 :HCVNS3丝氨酸蛋白酶与P53之间确实存在相互作用 ,但NS3参与相互作用的区段不在NS3的N端 ,而在其C端
OBJECTIVE: To investigate the relationship between HCV NS3 serine protease and P53 protein and lay a foundation for studying the role of NS3 serine protease in the pathogenesis of hepatocarcinogenesis induced by HCV. Methods: The recombinant plasmid of HCV NS3 serine protease gene and its N-terminal deletion mutant was constructed. The yeast two-hybrid system was used to qualitatively and quantitatively detect the interaction of P53 with NS3 and its co-mutant. Results: The HCV NS3 serine protease had a N-terminal deletion of 15 amino acids and a deletion of 30 amino acids of NS3 serine protease and P53 both interaction, and the intensity of interaction was not significantly different. CONCLUSION: There is indeed an interaction between HCV NS3 serine protease and P53, but the NS3-involved segment is not at the N-terminus of NS3, but at the C-terminus
