目的 探讨溶酶体中CathepsinB与扩张型心肌病发病的关系。 方法 收集我院因终末期扩张型心肌病行心脏移植术的受体标本 2 0例为实验组及脑死亡 3h内排除心脏疾患的心脏标本 5例为正常对照组 ,通过免疫组织化学 ,RT -PCR和WesternBlotting等方法自不同水平对CathepsinB进行检测。 结果 发现两组心肌组织中的CathepsinB均有阳性信号 ;RT PCR半定量检测CathepsinBmRNA的表达示扩张型心肌病组CathepsinB与 β actin条带面积积分比值明显高于对照组 ,分别为 0 .92 6± 0 .30、0 .4 4 2± 0 .2 5 ,两组间有显著差异 (P <0 .0 1) ;同时从蛋白水平检测CathepsinB的表达 ,显示扩张型心肌病组CathepsinB与 β actin条带面积积分比值明显高于正常组 ,分别为 0 .4 5 4± 0 .0 8及 0 .131± 0 .0 3,两组间有显著差异 (P <0 .0 1)。结论 与对照组相比 ,扩张型心肌病心肌标本中CathepsinBmRNA与蛋白质的表达量均明显增高 ,提示CathepsinB可能参与了扩张型心肌病的发生、发展过程 ,并推测其在扩张型心肌病心肌凋亡的过程中可能发挥了重要的作用
Objective To investigate the relationship between the lysosomal Cathepsin B and the development of dilated cardiomyopathy. Methods Twenty cases of cardiac transplant recipients with end-stage dilated cardiomyopathy in our hospital were selected as the experimental group and 5 cases of heart disease excluding heart disease within 3 hours after the brain death as normal control group. Immunohistochemistry, RT-PCR And WesternBlotting and other methods to detect CathepsinB from different levels. The results showed that there was a positive signal of CathepsinB in both groups. RT-PCR to detect the expression of CathepsinB mRNA showed that the ratio of Cathepsin B to β actin in dilation cardiomyopathy group was significantly higher than that of the control group (0.92 6 ± 0.30,0.42 ± 0.52, there was a significant difference between the two groups (P <0.01). At the same time, the expression of Cathepsin B was detected from the protein level, indicating that the expression of Cathepsin B and β actin in dilated cardiomyopathy group The area integral score was significantly higher than that of the normal group, which was respectively 0.45 ± 0.80 and 0.131 ± 0.30, with significant difference between the two groups (P <0.01). Conclusions Compared with the control group, the expression of Cathepsin B mRNA and protein in dilated cardiomyopathy specimens are significantly increased, suggesting that Cathepsin B may be involved in the development of dilated cardiomyopathy and its development, and presumably in the dilated cardiomyopathy myocardial apoptosis The process may have played an important role
