目的探讨thy-1与c-kit在肝癌中表达能否作为预测肝癌发生、发展的指标。方法免疫组化观察50例原发性肝癌及配对癌周组织中thy-1与c-kit的表达定位、与肝癌分化程度的关系及二者在肝癌中表达的相关性,ELISA观察53例原发性肝癌外周血中二者表达的相关性。结果 (1)免疫组织化学检测:thy-1与c-kit均表达在细胞质,染色强度从癌周组织-高分化-中低分化肝癌中呈逐步增强趋势,在癌周组织中表达显著低于在肝癌组织中表达,组间比较差异显著(χ2=17.761,27.656,P=0.000);二者在肝癌中表达与其分化程度有关,在高分化肝癌中表达均低于在中低分化肝癌中表达,组间比较差异显著(χ2=20.592,8.239,P=0.000,0.041)。thy-1与c-kit在肝癌中表达呈显著正相关(r=0.333,P=0.018),其中二者共阳性表达的42例肝癌标本中,高分化肝癌15例,中低分化肝癌27例,组间比较差异显著(χ2=10.258,P=0.023)。(2)ELISA检测示thy-1与c-kit在肝癌外周血中表达呈显著正相关(r=0.443,P=0.001)。结论 thy-1与c-kit在肝癌中的联合表达可作为肝癌发展及预后的指标。
Objective To investigate the expression of thy-1 and c-kit in hepatocellular carcinoma (HCC) as an index for predicting the occurrence and development of HCC. Methods Immunohistochemistry was used to observe the expression of thy-1 and c-kit in 50 primary hepatocellular carcinoma (HCC) and matched cancer tissues. The relationship between the expression of thy-1 and c-kit and the degree of differentiation of hepatocellular carcinoma was analyzed. Correlation of expression of the two in peripheral blood of patients with primary. Results (1) Immunohistochemical detection: Both thy-1 and c-kit were expressed in the cytoplasm. The staining intensity increased gradually from peritumoral tissue-well-differentiated-poorly-differentiated hepatocellular carcinoma and significantly lower than that in the peritumoral tissue (Χ2 = 17.761,27.656, P = 0.000). The expression of HCC and HCC in hepatocellular carcinoma was related to the degree of differentiation, and was lower in HCC than that in HCC , There was significant difference between the two groups (χ2 = 20.592, 8.239, P = 0.000,0.041). There was a significant positive correlation between the expression of thy-1 and c-kit in hepatocellular carcinoma (r = 0.333, P = 0.018). Of the 42 hepatocellular carcinomas with positive expression of thy-1, 15 were highly differentiated hepatocellular carcinoma, 27 were moderately poorly differentiated hepatocellular carcinoma , There was significant difference between the two groups (χ2 = 10.258, P = 0.023). (2) There was a significant positive correlation between the expression of thy-1 and c-kit in peripheral blood of HCC patients by ELISA (r = 0.443, P = 0.001). Conclusion The co-expression of thy-1 and c-kit in hepatocellular carcinoma can be used as an indicator of the development and prognosis of hepatocellular carcinoma.
