杂交瘤细胞的长期保存,一般都是冻存在液氮(或氮气)中,所用培基中含有胎牛血清(FCS)及二甲基亚砜(DMSO)。这种冻存杂交瘤细胞方法虽安全可靠,但需要液氮装置,并经常补充液氮,价值甚高。本文作者设计了一种以牛奶衍生物代替FCS作为冻存细胞的培基,置-80℃冰箱长期保存细胞的方法。牛奶衍生物为新鲜牛奶经过67℃加热处理,离心去脂肪、去细胞碎片的浓缩蛋白液。为了作对比,配制两种细胞冻存液,培基A含有40%牛奶衍生物和10%DMSO,培基B含有40%FCS及10%DMSO。实验用细胞是分泌抗人促性腺激素
