采用半固体软琼脂单层细胞培养法,测定4种口腔厌氧菌内毒素(LPS)诱导小鼠集落刺激因子(CSF)的活性。结果发现,0.1μg的内毒素即可诱导CSF增生,并随内毒素剂量增加而呈剂量依赖曲线。但当内毒素剂量超过50μg后,CSF水平反呈下降趋势。血清CSF产生高峰位于内毒素注射后6h。4种内毒素对CSF的诱导活性分别为65~88CFU—C。由内毒素诱生的CSF主要为诱导单核-巨噬细胞分化增殖的GM-CSF。
Semi-solid soft agar monolayer cell culture was used to measure the activity of 4 kinds of oral anaerobic bacteria endotoxin (LPS) induced mouse colony stimulating factor (CSF). The results showed that, 0.1μg of endotoxin can induce CSF proliferation, and dose-dependent endotoxin dose-dependent manner. However, when the dose of endotoxin exceeds 50μg, the level of CSF shows a downward trend. The peak of serum CSF production is located 6 h after endotoxin injection. The inducing activities of four kinds of endotoxin on CSF were 65-88CFU-C respectively. CSF induced by endotoxin is mainly GM-CSF that induces monocyte-macrophage differentiation and proliferation.
