目的探讨女性绝经后骨质疏松症患者雌激素受体(ER)基因Px单倍型对骨密度的影响。方法采用聚合酶链反应方法检测197例患者ER基因Xba I和PvuⅡ多态性,双能X线骨密度仪检测腰椎和股骨上端的骨密度。结果基因型分布:XX、xx、Xx、PP、pp、Pp型分别为18.8%、13.2%、68%、19.3%、34%、46.7%;Xba I、PvuⅡ单个多态位点与骨密度无相关性(P>0.05),2个位点联合有Px单倍型组Ward’s区骨密度(0.47±0.10)g/cm2明显低于无Px单倍型组(0.51±0.13) g/cm2(P<0.05);影响腰椎骨密度的自身主要因素:有Px单倍型组为体重,无Px单倍型组为身高。结论ER基因Px单倍型可能对绝经后骨质疏松症骨密度有不利影响,Px单倍型有助于临床判断身高和体重对骨密度的影响。
Objective To investigate the effects of Px haplotype of estrogen receptor (ER) gene on bone mineral density in female patients with postmenopausal osteoporosis. Methods The genotypes of Xba I and Pvu Ⅱ of ER gene in 197 patients were detected by polymerase chain reaction. Bone mineral density (BMD) in lumbar vertebrae and femur was measured by dual energy X-ray absorptiometry. Results The genotypes of genotypes were 18.8%, 13.2%, 68%, 19.3%, 34% and 46.7% in XX, XX, Xx, PP, There was no correlation between polymorphism sites and bone mineral density (P> 0.05). The bone density (0.47 ± 0.10) g / cm2 in Ward’s area at 2 sites combined with Px haplotype was significantly lower than that without Px Haplotype group (0.51 ± 0.13) g / cm2 (P <0.05). The main factors affecting lumbar BMD were Px haplotype group and no Px haplotype group . Conclusion The Px haplotype of ER gene may have an adverse effect on the bone mineral density of postmenopausal osteoporosis. The Px haplotype may help to determine the effect of height and weight on the bone mineral density.
