目的:首次建立同时测定毛冬青中Ilexsaponin B1、Ilexsaponin B2和Ilexgenin A含量的方法并将其运用到同属植物三萜的检测中。方法:采用Cemini C18色谱柱(4.6 mm×250 mm,5μm),流动相为乙腈-0.5%醋酸水,流速1.0 mL/min,蒸发光散射检测器:漂移管温度为107℃,气体流速为3.0 L/min。结果:Ilexsaponin B1、Ilexsaponin B2和Ilexgenin A线性范围分别为0.25~2.50μg(r1=0.9992)、0.03~0.30μg(r2=0.9995)和0.07~0.70μg(r3=0.9998),平均回收率(n=6)分别为101.08%、100.13%和100.16%。毛冬青及其同属植物中3种五环三萜Ilexsaponin B1、Ilexsaponin B2和Ilexgenin A的含量差异较大,其中在毛冬青中的含量最高。结论:该方法操作准确、有效,可为冬青属药材的来源和开发利用提供依据,同时测定3种五环三萜的方法可用于冬青属特别是毛冬青药材的质量控制。
OBJECTIVE: To establish a method for the simultaneous determination of Ilexsaponin B1, Ilexsaponin B2 and Ilexgenin A in Mao Dongqing and to apply it to the detection of the triterpenoid in the same genus. Methods: Cemini C18 column (4.6 mm × 250 mm, 5 μm) was used. The mobile phase consisted of acetonitrile-0.5% acetic acid and the flow rate was 1.0 mL / min. The evaporative light scattering detector was used. The drift tube temperature was 107 ℃ and the gas flow rate was 3.0 L / min. Results: The linear ranges of Ilexsaponin B1, Ilexsaponin B2 and Ilexgenin A were 0.25-2.50μg (r1 = 0.9992), 0.03-0.30μg (r2 = 0.9995) and 0.07-0.70μg (r3 = 0.9998) 6) were 101.08%, 100.13% and 100.16% respectively. The contents of three kinds of pentacyclic triterpenes, Ilexsaponin B1, Ilexsaponin B2 and Ilexgenin A in Mao Dongqing and its related genus were significantly different, among them the content was the highest in Mao Dongqing. Conclusion: The method is accurate and effective. It can provide the basis for the origin and development and utilization of the medicinal materials of Holly, and the determination of three kinds of pentacyclic triterpenoids can be used for the quality control of Holly, especially Mao Dongqing.
