目的探讨突触核蛋白-γ(SNCG)表达对乳腺癌雌激素受体(ER)表达的影响。方法构建慢病毒SNCG过表达及干扰载体,分别转染人乳腺癌T47D(SNCG+/ER+)细胞株。在SNCG过表达和SNCG干扰细胞组中,RT-PCR方法检测SNCG和ER的mRNA表达水平,Westernblot法检测SNCG和ER的蛋白表达水平,并做Pearson法相关统计分析。结果在mRNA水平,SNCG过表达组中ER基因的mRNA表达量显著增高,两者呈正相关(r=0.81,P=0.03);SNCG干扰组中ER基因的mRNA表达量显著降低,两者呈正相关(r=0.82,P=0.01)。在蛋白表达水平,SNCG过表达组ER表达显著增强,两者呈正相关(r=0.85,P=0.03);SNCG干扰组ER表达显著降低,两者呈正相关(r=0.83,P=0.04)。结论 SNCG对ER表达有正向调节作用,为进一步研究SNCG与ER的相互作用机制及SNCG与乳腺癌内分泌治疗之间的关系奠定实验基础。
Objective To investigate the effect of synuclein-γ (SNCG) expression on the expression of estrogen receptor (ER) in breast cancer. Methods The lentivirus SNCG overexpression and interference vector was constructed and transfected into human breast cancer T47D (SNCG + / ER +) cell lines respectively. In SNCG overexpression and SNCG interfering cell groups, the mRNA expression levels of SNCG and ER were detected by RT-PCR. The protein expression levels of SNCG and ER were detected by Western blot and analyzed by Pearson correlation. Results At the mRNA level, the expression of ER mRNA in SNCG overexpression group was significantly increased (r = 0.81, P = 0.03). The mRNA expression of ER gene in SNCG interference group was significantly decreased (r = 0.82, P = 0.01). The expression of ER in SNCG overexpression group was significantly increased at the level of protein expression (r = 0.85, P = 0.03). The ER expression was significantly decreased in SNCG interference group (r = 0.83, P = 0.04). Conclusion SNCG has a positive regulatory effect on ER expression, which lays the foundation for further study on the mechanism of SNCG interaction with ER and the relationship between SNCG and endocrine therapy of breast cancer.
