目的:研究补肾益精法中药复方对硬皮病小鼠皮肤纤维化表型的影响。方法:使用博来霉素诱导建立硬皮病小鼠模型,分别采用补肾益精法中药复方(中药复方组)、生理盐水(模型组)处理硬皮病模型小鼠,共3周,并设正常对照组。HE染色观察皮肤病理改变;使用实时定量逆转录-聚合酶链反应(RT-PCR)技术,测定各组小鼠I型胶原α2(COLⅠα2)基因表达情况。结果:成功建立硬皮病小鼠模型,皮肤病理HE染色显示,中药复方组小鼠皮肤纤维化有不同程度改善。模型组小鼠COLⅠα2基因表达较正常对照组高(P<0.01);经补肾益精法处理后,中药复方组小鼠COLⅠα2基因表达减少,与正常对照组比较,差异无显著性意义(P>0.05)。结论:补肾益精法中药复方可改善硬皮病模型小鼠皮肤纤维化程度。
Objective: To study the effect of Bushen Yijing method on skin fibrosis phenotype in scleroderma mice. Methods: A mouse model of scleroderma was induced by bleomycin. The scleroderma mice were treated with Bushen Yijing Compound (TCM compound) and saline (model group) for 3 weeks respectively Normal control group. The pathological changes of the skin were observed by HE staining. The expression of collagen Ⅰ 2 (COLⅠα2) gene in each group was measured by real-time quantitative reverse transcription-polymerase chain reaction (RT-PCR). Results: The mouse model of scleroderma was successfully established. HE staining showed that skin fibrosis of the compound prescription group mice improved to some extent. Compared with normal control group, the expression of COLⅠα2 in model group mice was higher than that in normal control group (P <0.01). After treated by Bushenyijing method, the expression of COLⅠα2 gene in mice of compound traditional Chinese medicine group decreased, compared with normal control group, there was no significant difference (P> 0.05). Conclusion: Bushen Yijing Pharmacotherapy can improve the degree of skin fibrosis in scleroderma mice.
