为明确黑米种皮色素的遗传基础,利用籼稻黑米黑B和无色东北粳稻杂交,获得F1和F2分离群体。F1种皮表现黑色,说明种皮中的黑色性状受显性基因控制。同时,其F2代后代表现3:1的孟德尔分离比,表明黑色性状基因是受一对显性基因控制,暂定名为BP(black pericarp)。利用F2分离群体作为定位群体,用均匀分布在水稻12条染色体上的128对SSR引物和45对indel引物进行BSA和分子标记分析,利用MAPMAKER3.0软件进行连锁分析,将控制黑色性状基因定位在第4染色体上。目的基因BP位于引物R4M23和R4M43之间,遗传距离分别为10.2c M和14.6c M。
In order to clarify the genetic basis of pigment in seed coat of black rice, F1 and F2 segregation populations were obtained by crossing indica black rice black B and colorless northeast japonica rice. The F1 seed coat showed black color, indicating that the black trait in the seed coat is controlled by the dominant gene. At the same time, the F2 generation showed a 3: 1 Mendelian segregation ratio, indicating that the black trait gene is controlled by a pair of dominant genes and tentatively named BP (black pericarp). Using F2 segregating population as the locating population, 128 pairs of SSR primers and 45 pairs of indel primers uniformly distributed on 12 chromosomes of rice were analyzed by BSA and molecular markers. MAPMAKER3.0 software was used for linkage analysis, On chromosome 4. The target gene BP was located between the primers R4M23 and R4M43 with genetic distances of 10.2c M and 14.6cM, respectively.
