作者对正常大鼠、卵巢切除大鼠、卵巢切除术后给与不同剂量雌激素大鼠的心房肽基因表达情况进行了研究。用改良冷酚法提取鼠心房总RNA,用α-~(32)p标记的与ANP mRNA互补的cDNA探针与RNA样品杂交。分子杂交结果显示:OVX组ANP含量相对最低,其对照E-20组、E-200组和假手术组的含量分别是其1.3、1.7和1.1倍。此结果提示,雌激素促进ANP的基因表达;进一步说,这种增强作用与雌激素的使用剂量有关。生理剂量的雌激素具有维持ANP适当转录水平的作用。
The authors studied the atrial natriuretic peptide gene expression in normal rats, ovariectomized rats and ovariectomized rats with different doses of estrogen. Total RNA of rat atrium was extracted by modified cold phenol method, and RNA probes were hybridized with cDNA probes complementary to α - (32) p labeled with ANP mRNA. The result of molecular hybridization showed that the content of ANP in OVX group was the lowest, the contents of ANP in E-20 group, E-200 group and sham-operated group were 1.3, 1.7 and 1.1 times respectively. This result suggests that estrogen promotes gene expression of ANP; further, this enhancement is related to the dose of estrogen used. Physiological doses of estrogen have the effect of maintaining an appropriate level of ANP transcription.
