目的 研究房水的免疫抑制功能,分析转化生长因子-β2(TGF-β2)在其中的作用。方法 分别用正常房水、酸化房水和加有TGF-β2抗体的酸化房水作用于淋巴细胞,通过 MTT比色法测定各种处理因素下的房水对脐带血淋巴细胞增殖的抑制作用,并计算抑制率。结果 正常房水可抑制淋巴细胞的增殖,抑制率为7. 87%;酸化房水的抑制作用明显加强,抑制率提高到 51. 53%;TGF-β2抗体可阻断酸化房水的活性,加入TGF-β2抗体后,抑制率下降到 9. 7%。结论 房水具有免疫抑制功能,可抑制淋巴细胞的增殖,而TGF-β2是产生这一作用的分子学基础。
Objective To study the immunosuppressive function of aqueous humor and to analyze the role of transforming growth factor-β2 (TGF-β2) in it. Methods The normal aqueous humor, acidified aqueous humor and acidified aqueous humor with TGF-β2 antibody were respectively used in lymphocytes. MTT assay was used to determine the inhibitory effect of aqueous humor on the proliferation of umbilical cord blood lymphocytes under various treatment conditions. And calculate the inhibition rate. Results Normal aqueous humor inhibited the proliferation of lymphocytes, the inhibition rate was 7. 87%; inhibition of acidified aqueous humor significantly enhanced inhibition rate increased to 51.53%; TGF-β2 antibody can block acidified aqueous humor activity, 7%. After adding TGF-β2 antibody, the inhibition rate dropped to 9.7%. Conclusion Aqueous humor has immunosuppressive function, which can inhibit the proliferation of lymphocytes. TGF-β2 is the molecular basis for this effect.
