本研究测定了兔胚泡肽(RBP2)的氨基酸序列,它由54个氨基酸组成,其序列的第4位氨基酸至第34位氨基酸与兔子宫珠蛋白N端氨基酸序列完全相同。用计算机软件系统模拟分析,从54个氨基酸中选取了抗原性最高的一段氨基酸(aa20-aa34),然后人工合成了含有15个氨基酸的小肽片段(SPF)。研究表明:用溴脱氧尿嘧啶掺入淋巴细胞的方法测定了SPF对细胞增殖作用的影响,当浓度范围在80μg/ml至320μg/ml时,SPF的免疫抑制作用不明显。与天然RBP2比较,后者有明显的抑制作用。此结果说明,SPF的免疫抑制效应明显低于RBP2。实验又证明SPF(浓度范围在45~180μg/ml培液)对兔子宫内膜PGF2α的分泌呈剂量相关的抑制作用。此外,用[3H]亮氨酸掺入法,证明SPF(浓度180μg/ml培波)对[3H]亮氨酸掺入子宫内膜有明显促进作用,此促进作用被蛋白质抑制剂放线菌素(achdine)所阻断。
In this study, we determined the amino acid sequence of rabbit blastocyst (RBP2) peptide, which consists of 54 amino acids. The amino acid sequence from amino acid 4 to amino acid 34 of rabbit RBG2 is exactly the same as the N-terminal amino acid sequence of rabbit uteroglobin. A computer software system was used to select aa20-aa34 with the highest antigenicity from 54 amino acids. Then, a small peptide fragment (SPF) containing 15 amino acids was synthesized. The results showed that the effect of SPF on the cell proliferation was measured by the incorporation of bromodeoxyuridine into lymphocytes. The immunosuppressive effect of SPF was not obvious when the concentration ranged from 80μg / ml to 320μg / ml. Compared with natural RBP2, the latter has significant inhibitory effect. This result shows that the immunosuppressive effect of SPF is significantly lower than that of RBP2. The experiment also proved SPF (concentration in the range of 45 ~ 180μg / ml) on the secretion of endometrial PGF2α in a dose-dependent manner. In addition, the incorporation of [3H] leucine showed that SPF (180μg / ml) significantly enhanced the incorporation of [3H] leucine into the endometrium, which was enhanced by the protein inhibitor actinomycetes Blocked by achdine.
