目的研究高乌头炮制前后水煎液和三氯甲烷部分对小鼠镇痛抗炎作用。方法取SPF级昆明小鼠,按照体重随机分为10组:空白对照组(0.5%甲基纤维素)、阳性对照组(消炎痛10 mg·kg~(-1),氢溴酸高乌甲素1.6 mg·kg~(-1))、高乌头水煎液高低2个剂量组(0.9,0.3g·kg~(-1)),制高乌头水煎液高低2个剂量组(0.9,0.3 g·kg~(-1)),高乌头三氯甲烷高低2个剂量组(4.5,1.5 mg·kg~(-1)),制高乌头三氯甲烷高低2个剂量组(4.5,1.5 mg·kg~(-1)),每组10只。采用热板法对高乌头炮制前后水煎液及三氯甲烷部分进行镇痛作用评价;用二甲苯致鼠耳肿胀实验对其进行抗炎作用评价。结果镇痛作用:在热板法实验中,给药后30 min,制品水煎液组小鼠痛阈值为(24.1±6.3)s;给药后120 min后,制品水煎液组痛阈值为(49.7±9.7)s,生品三氯甲烷部分高低2个剂量组、制品三氯甲烷部分高剂量组痛阈值分别为(49.8±7.4),(47.3±6.3),(45.1±7.1)s,与空白对照组相比,差异具有统计学意义(均P<0.05),说明高乌头的镇痛作用发生在给药后120 min左右。抗炎作用:高乌头生品水煎液组、制品水煎液高低2个剂量组、高乌头生品和制品三氯甲烷部分高低2个剂量组小鼠耳肿胀度分别为(16.9±11.2),(15.2±4.8),(17.7±6.4),(14.2±4.2),(16.0±4.5),(15.7±5.2),(12.5±5.5)mg,与空白对照组相比,差异具有统计学意义(均P<0.01)。制品各组与相应生品各组比较,差异无统计学意义(P>0.05)。结论乌头生品具有很强的镇痛抗炎活性,炮制后其镇痛抗炎活性仍然保持,并有增强的趋势。
Objective To study the analgesic and anti-inflammatory effects of decoction and trichloromethane fraction before and after high-aconite processing. Methods SPF Kunming mice were randomly divided into 10 groups according to body weight: blank control group (0.5% methylcellulose), positive control group (indomethacin 10 mg · kg -1, (1.6 mg · kg -1), 2 doses of high aconite decoction (0.9,0.3 g · kg -1) and 2 doses of high aconite decoction 0.9,0.3 g · kg -1) and high and low aconitum chloroform (2 groups, 4.5 and 1.5 mg · kg -1) (4.5, 1.5 mg · kg -1), 10 in each group. The hot plate method was used to evaluate the analgesic effect of the decoction and the chloroform fraction before and after the treatment of A. capitatum. The anti-inflammatory effect was evaluated using xylene-induced mouse ear swelling test. Results The analgesic effect: In the hot plate test, the pain threshold value of the decoction group was (24.1 ± 6.3) s at 30 min after administration, and the pain threshold of the decoction group was (49.7 ± 9.7) s, two parts of high and low levels of chloroform, the pain thresholds of the high dose of chloroform were (49.8 ± 7.4), (47.3 ± 6.3) and (45.1 ± 7.1) s, Compared with the blank control group, the difference was statistically significant (all P <0.05), indicating that the analgesic effect of high aconite occurs after about 120 min. Anti-inflammatory effects: high aconite root decoction group, the level of decoction decoction 2 doses group, high aconite raw materials and products part of the level of chloroform trichloride two doses of mice ear swelling degree were (16.9 ± 11.2), (15.2 ± 4.8), (17.7 ± 6.4), (14.2 ± 4.2), (16.0 ± 4.5), (15.7 ± 5.2) and (12.5 ± 5.5) mg, respectively. Compared with the blank control group, Significance (both P <0.01). There was no significant difference between each group and the corresponding product (P> 0.05). Conclusion Aconitum has a strong analgesic and anti-inflammatory activity, analgesic and anti-inflammatory activity after treatment still maintained, and there is an increasing trend.
