目的 探讨雷公藤内酯醇对小鼠脾淋巴细胞IL 2产生和IL 2受体表达的影响。方法应用体外细胞培养和APAAP桥联酶标法检测了雷公藤内酯醇对刀豆蛋白A(ConA)诱导的小鼠脾淋巴细胞白细胞介素 (IL -2 )产生及IL -2受体表达的影响。结果雷公藤内酯醇对IL -2产生无影响 ,但对IL -2受体表达有明显的抑制作用。结论雷公藤内酯醇抗免疫排斥作用主要是通过抑制IL -2受体表达 ,从而使IL -2与IL -2受体的相互作用受到影响 ,最终抑制了淋巴细胞增殖。
Objective To investigate the effects of triptolide on IL 2 production and IL 2 receptor expression in splenic lymphocytes of mice. Methods The effects of triptolide on IL-2 production and IL-2 receptor expression in mouse splenic lymphocytes induced by ConA were studied by in vitro cell culture and APAAP linked enzyme assay influences. Results Triptolide had no effect on IL-2 production, but had significant inhibitory effect on IL-2 receptor expression. CONCLUSION: The anti-immune rejection of triptolide mainly inhibits the expression of IL-2 receptor, which affects the interaction between IL-2 and IL-2 receptor and eventually inhibits the proliferation of lymphocytes.
