目的:在提纯兔视网膜S抗原的基础上,进一步观察其蛋白质构象,为深入研究该抗原的免疫性打下基础。方法:利用二步离子交换法及高效液相层析法提纯兔视网膜S抗原,采用隧道扫描电子显微镜对S抗原的构象进行分析观察。结果:对S抗原的空间构象进行观察的结果发现,兔视网膜S抗原呈椭圆球状构象,其二维尺寸为7.5nm×5.3nm(X轴×Y轴),与其分子量55000十分相符。确定了S抗原为球蛋白结构。讨论:从我们掌握的文献来看,利用隧道扫描电镜观察S抗原的构象并进行分析,在国内外尚属首次。实验结果表明,S抗原为球蛋白构象。隧道扫描电子显微镜对研究生物样品十分有利,可以使我们观察到更接近生理条件下的分子构象,为研究抗原决定簇,进而研究抗原免疫性提供良好的手段。
OBJECTIVE: To further observe the protein conformation on the basis of purifying the rabbit retina S antigen and lay a foundation for further study of the antigen immunity. Methods: Rabbit retinal S antigen was purified by two-step ion-exchange method and high-performance liquid chromatography. The conformation of S antigen was observed by tunnel scanning electron microscopy. Results: Observing the spatial conformation of S antigen, the rabbit retina S antigen showed an ellipsoidal conformation with a 2D dimension of 7.5 nm × 5.3 nm (X-axis × Y-axis), which was in good agreement with its molecular weight of 55,000. S antigen was identified as globulin structure. Discussion: From the literature we have, it is the first time to observe the conformation of S antigen by tunnel scanning electron microscope and analyze it. The experimental results show that S antigen is globulin conformation. Tunnel scanning electron microscopy is very beneficial for the study of biological samples, allowing us to observe the molecular conformation closer to physiological conditions, providing a good means for the study of antigenic determinants and further study of antigenic immunity.
