目的通过检测胃肠道肿瘤中CXCR4的作用方式来探讨在发病时它的作用。方法通过逆转录+Western blot和即时定量PCR的方法分别来检测CXCR4在胃肠道肿瘤的细胞系和标本中的表达。结果结直肠癌组织标本和胃癌组织标本中CXCR4 mRNA的表达水平要比其对应的配对癌旁正常组织的高。胃肠道肿瘤细胞系SW-480、HT-29、AGS和SGC-7901中都存在有CXCR4mRNA和CXCR4蛋白的表达。CXCR4的表达和胃癌患者淋巴结转移及分期状态具有相关性。结论在胃肠道肿瘤中CXCR4表达过度并且参与了发病过程。
Objective To explore the role of CXCR4 in detecting gastrointestinal cancers at onset. Methods The expression of CXCR4 in gastrointestinal tumor cell lines and specimens were detected by reverse transcription + Western blot and real-time quantitative PCR, respectively. Results The expression level of CXCR4 mRNA in colorectal cancer tissue specimens and gastric cancer specimens was higher than that in corresponding paired normal tissues. The expression of CXCR4 mRNA and CXCR4 protein is present in the gastrointestinal tumor cell lines SW-480, HT-29, AGS and SGC-7901. The expression of CXCR4 is correlated with lymph node metastasis and staging status in patients with gastric cancer. Conclusion CXCR4 is overexpressed and involved in the pathogenesis of gastrointestinal cancers.
