氨苄青霉素在人体内代谢产物已知有5R,6R-青霉噻唑酸和5S,6R-差相异构体。近年来,又在大白鼠尿中发现哌嗪-2,5-二酮(P-2,5d)为氨苄青霉素的一种新代谢产物。本文报道采用HPLC测定口服氨苄青霉素后人尿液中的P-2,5-d。P-2,5-d和5×10~(-4)M的最适浓度的氯化亚汞在pH7.0反应,在305nm处有最大紫外吸收,反应产物稳定;如氯化亚汞过量则紫外吸收消失。反应产物虽然在酸性pH范围(pH 1.8~3.6)比在中性pH范围的稳定性
Ampicillin is known as 5R, 6R-penicillin and 5S, 6R-epimers in human metabolites. In recent years, piperazine-2,5-dione (P-2,5d) has also been found in rat urine as a new metabolite of ampicillin. This article reports the determination of P-2,5-d in human urine by oral ampicillin using HPLC. The optimal concentrations of P-2,5-d and 5 × 10 -4 M for the reaction of mercuric chloride with pH7.0 and the maximum UV-absorbance at 305 nm resulted in the stabilization of the reaction product. For example, excess mercuric chloride UV absorption disappears. The reaction product is stable in the acidic pH range (pH 1.8 to 3.6) than in the neutral pH range
