目的检测牙龈卟啉单胞菌菌毛不同fimA基因型在颈动脉粥样硬化斑块中的分布。方法选取需要进行颈动脉内膜剥脱术的患者47例,收集术中分离的颈动脉粥样硬化斑块和患者龈下菌斑,用16S rRNA PCR检测牙龈卟啉单胞菌菌毛不同fimA基因型的分布。结果在47例患者中,牙龈卟啉单胞菌在龈下菌斑和颈动脉粥样硬化斑块中的检出率分别为72.3%和46.8%,在同一患者同时检出率为40.4%。Type Ⅱ fimA在龈下菌斑和颈动脉粥样硬化斑块中的检出率最高,分别为76.4%和81.8%;其次为Type Ⅳ fimA,检出率分别为50%和40.9%;这两种基因型的检出率显著高于其他基因型。牙龈卟啉单胞菌Type Ⅴ fimA基因型在龈下菌斑和动脉粥样硬化斑块中均未检出。结论牙龈卟啉单胞菌及其不同fimA基因型同时在龈下菌斑和颈动脉粥样硬化斑块中被检出,Type Ⅱ和Ⅳ fimA基因型牙龈卟啉单胞菌作为主要的定植菌,可能在颈动脉粥样硬化发生和发展中起到了一定的作用。
Objective To detect the distribution of different fimA genotypes of Porphyromonas gingivalis in carotid atherosclerotic plaques. Methods Forty-seven patients with carotid endarterectomy were enrolled in this study. Carotid artery atherosclerotic plaques and subgingival plaque were collected. The different fimA genes of Porphyromonas gingivalis pilus were detected by 16S rRNA PCR Type of distribution. Results In 47 patients, the detection rates of Porphyromonas gingivalis in subgingival plaques and carotid atherosclerotic plaques were 72.3% and 46.8%, respectively, while the same patient was detected in 40.4% at the same time. Type Ⅱ fimA had the highest detection rate in subgingival plaques and carotid atherosclerotic plaques, which were 76.4% and 81.8% respectively, followed by Type Ⅳ fimA with the detection rates of 50% and 40.9% respectively The detection rate of genotypes was significantly higher than other genotypes. Porphyromonas gingivalis Type Ⅴ fimA genotype in subgingival plaque and atherosclerotic plaques were not detected. Conclusion Porphyromonas gingivalis and its different fimA genotypes were detected in both subgingival plaque and carotid atherosclerotic plaques. Type Ⅱ and Ⅳ fimA genotypes of Porphyromonas gingivalis were the main colonizing bacteria , May play a role in the occurrence and development of carotid atherosclerosis.
