目的 探讨视神经损伤后 ,信号转导子与转录激活子 (STAT3)蛋白在视网膜内表达和分布的变化。方法 免疫细胞化学、Westernblot及计算机图像分析技术。 结果 视神经切断术后 ,视网膜内STAT3表达水平上调 ,术后 1d达到高峰 ,并出现核转位增多现象。 5d后STAT3水平逐渐下降至正常。 结论 Janus激酶 信号转导子与转录激活子 (JAK STAT)信号转导途径可能通过STAT3蛋白的活化 ,参与视神经切断后视网膜内的病理改变过程
Objective To investigate the expression and distribution of signal transducer and activator of transcription 3 (STAT3) protein in the retina after optic nerve injury. Methods Immunocytochemistry, Western blot and computer image analysis techniques. Results After optic nerve transection, the level of STAT3 in the retina was up-regulated and the peak was reached on the 1st day after operation. The phenomenon of increased nuclear translocation appeared. After 5d STAT3 level gradually decreased to normal. Conclusions Janus kinase signal transducer and activator of transcription (JAK STAT) signal transduction pathway may be involved in the process of pathological changes in the retina after optic nerve transection through the activation of STAT3 protein
