目的分析福建省鼠伤寒及肠炎沙门菌脉冲场凝胶电泳(PFGE)指纹图谱,研究其分型特征。方法收集2009年50株沙门菌临床分离株,其中30株鼠伤寒沙门菌分离自婴幼儿腹泻患者,20株肠炎沙门菌分离自感染性腹泻患者。选择XbaⅠ酶对沙门菌染色体DNA进行酶切,H9812作为脉冲场凝胶分子量标准。采用PFGE技术分析电泳酶切指纹图谱,运用BioNumerics软件进行聚类分析。结果 30株婴幼儿腹泻鼠伤寒沙门菌PFGE图谱按照100%的相似度可分为19个PFGE型,P1(Typhi)型有8株,P12(Typhi)型3株,P1(Typhi)为优势型别;20株肠炎沙门菌PFGE图谱按照100%的相似度可分为10个PFGE型,其中P1(Enteri)型6株,P4(Enteri)型有4株,P6(Enteri)型3株,P1(Enteri)型为优势型别。结论 PFGE分型方法分辨率高,重复性好,可广泛运用于沙门菌疫情溯源和早期防控。
Objective To analyze the fingerprint of Salmonella typhimurium pulsed-field gel electrophoresis (PFGE) in Fujian Province and study its classification characteristics. Methods Fifty clinical isolates of Salmonella were collected in 2009, of which 30 Salmonella typhimurium isolates were isolated from infantile diarrhea patients and 20 Salmonella enteritidis isolated from patients with infectious diarrhea. Select Xba Ⅰ enzyme to digest the DNA of Salmonella chromosome, H9812 as the pulse field gel molecular weight standard. PFGE was used to analyze electrophoresis fingerprinting, and BioNumerics software was used for cluster analysis. Results The PFGE patterns of Salmonella typhimurium in 30 infants were divided into 19 PFGE patterns according to 100% similarity. There were 8 strains of P1 (Typhi) type, 3 strains of P12 (Typhi) type and P1 (Typhi) The 20 PFGE patterns of Salmonella enteritidis were classified according to 100% similarity, including 6 strains of P1 (Enteri), 4 strains of Enteri (P4), 3 of P6 (Enteri) and P1 (Enteri) type is the dominant type. Conclusion The PFGE typing method has high resolution and good repeatability and can be widely used in the traceability and early prevention and control of Salmonella.
