目的探讨经腹腔注射锰后,大鼠大脑核受体相关因子1(Nurr1)的改变。方法健康雄性2月龄SD大鼠50只,按体重随机分为5组:生理盐水组(A)、Mn2+2.5 mg/kg(B)、Mn2+5 mg/kg(C)、Mn2+10 mg/kg(D)、Mn2+20mg/kg(E),每天腹腔注射0.5 ml锰溶液,连续染毒30 d。实时荧光定量PCR检测大鼠脑组织中Nurr1 mRNA的相对表达水平,Western blot检测Nurr1的蛋白表达水平。结果脑组织Nurr1 mRNA相对表达的结果显示,与A组相比,B、C、D组Nurr1 mRNA含量降低,分别是A组的0.85,0.66,0.54和0.33倍,经多组比较的单因素方差分析显示不同浓度组间差异有统计学意义(P<0.01)。同时,脑组织Nurr1蛋白含量也随染锰剂量增大而降低,经多组比较的单因素方差分析显示差异有统计学意义(P<0.01)。结论锰可以在mRNA和蛋白质水平改变Nurr1的表达。
Objective To investigate the changes of nuclear receptor related factor 1 (Nurr1) in rat brain after intraperitoneal injection of manganese. Methods Fifty male 2-month-old male Sprague-Dawley rats were randomly divided into 5 groups according to body weight: saline (A), Mn2 + 2.5 mg / kg (B), Mn2 + 5 mg / mg / kg (D) and Mn2 + 20 mg / kg (E) were intraperitoneally injected with 0.5 ml of manganese solution for 30 days. The relative expression of Nurr1 mRNA in rat brain was detected by real-time fluorescence quantitative PCR, and the protein expression of Nurr1 was detected by Western blot. Results The relative expression of Nurr1 mRNA in brain tissue showed that compared with group A, the levels of Nurr1 mRNA in group B, C and D were decreased, which were 0.85, 0.66, 0.54 and 0.33 times respectively in group A, Analysis showed that there was a significant difference between different concentration groups (P <0.01). At the same time, the content of Nurr1 protein in brain tissue also decreased with the increase of manganese dose. The one-way ANOVA showed that the difference was statistically significant (P <0.01). Conclusion Manganese can change the expression of Nurr1 at mRNA and protein levels.
