目的观察右旋柠烯(D-Limonene)对大鼠大肠癌淋巴结转移及淋巴管生成的影响,为临床大肠癌术后治疗提供理论支持。方法二甲基肼诱发大鼠大肠癌模型40例。14周将大鼠分为治疗组和对照组,前者给予5%D-limonene(色拉油稀释)灌胃,后者给予等量色拉油做阴性对照。28周取材,免疫组化染色LYVE-1标记淋巴管,观察其分布。结果 LYVE-1标记的淋巴管多分布在癌周组织,中心区域未见淋巴管的阳性表达,D-limonene治疗组淋巴结转移率为5%,淋巴管数密度为2.4±0.83,与对照组(40%,6.1±1.35)比较,差异均有统计学意义(P<0.05)。结论 D-limonene减少肿瘤组织淋巴管的生成,降低淋巴道的转移。
Objective To observe the effect of D-Limonene on lymph node metastasis and lymphangiogenesis of colorectal cancer in rats and provide theoretical support for the treatment of colorectal cancer after operation. Methods 40 cases of rat model of colorectal cancer induced by dimethylhydrazine. At 14 weeks, the rats were divided into treatment group and control group. The former was given intragastric administration of 5% D-limonene (the salad oil was diluted) and the latter was given the same amount of salad oil as the negative control. Twenty-eight weeks later, LYVE-1 was labeled by immunohistochemistry to observe the distribution of lymphatic vessels. Results LYVE-1-labeled lymphatics were distributed in the peritumoral tissues and no lymphatic vessels were positive in the central area. Lymph node metastasis rate was 5% in D-limonene group and 2.4 ± 0.83 in lymphatic vessels. Compared with control group 40%, 6.1 ± 1.35), the differences were statistically significant (P <0.05). Conclusion D-limonene reduces lymphangiogenesis in tumor tissue and reduces lymph node metastasis.
