在抗Thy1.1抗体引起的大鼠系膜增殖性肾炎中,肾小球的增殖性细胞核抗原(prolifer-atingcellnuclearantigen,PCNA)阳性细胞增多, ̄3H-胸腺嘧啶核苷( ̄3H-thymidine, ̄3H-TdR)掺入法测定的肾小球增殖活性(glomerularproliferativeactivity,GPA)升高。对肾炎鼠给予5-脂氧合酶抑制剂MK886或血栓素(thromboxaneA2,TXA2)合成酶抑制剂Furegrelate或12-脂氧合酶抑制剂Baicalein进行处理,可分别抑制肾小球合成的白三烯(leukotricne,LT)B4 及12-羟二十碳四烯酸(12-hydroxyeicosatetraenoicacid,12-HETE),同时也抑制PCNA(+)细胞及GPA的上升。用X线照射或抗血小板抗体分别去除肾炎鼠的循环白细胞或血小板,均可抑制PCNA(+)细胞及GPA的上升。结果提示白细胞及血小板衍生的LT、TXA_2及12-HETE参与本模型的系膜细胞增殖。
Thy1.1 antibody-induced rat mesangial proliferative glomerulonephritis, glomerular proliferative cell nuclear antigen (prolifer-atingcellnuclearantigen, PCNA) positive cells, ~ 3H-thymidine (~ 3H-thymidine, ~ 3H-TdR incorporation assay showed increased glomerularproliferative activity (GPA). Nephritis mice were treated with 5-lipoxygenase inhibitor MK886 or thromboxane A2 (TXA2) synthase inhibitor Furegrelate or 12-lipoxygenase inhibitor Baicalein for the treatment of glomerular synthesis of leukotrienes Leukotricne (LT) B4 and 12-hydroxyeicosatetraenoic acid (12-HETE) as well as the increase of PCNA (+) cells and GPA. Using X-ray irradiation or anti-platelet antibody to remove circulating white blood cells or platelets of nephritic rats, respectively, can inhibit the increase of PCNA (+) cells and GPA. The results suggest that leukocytes and platelet-derived LT, TXA 2 and 12-HETE are involved in mesangial cell proliferation in this model.
