目的探讨白藜芦醇对卵巢癌细胞生长及细胞周期的影响,并研究白藜芦醇诱导卵巢癌细胞凋亡及其途径。方法MTT比色法检测白藜芦醇对细胞生长的影响,Ho33342染色荧光显微镜观察白藜芦醇对细胞核浓缩及片段化的影响,TUNEL检测白藜芦醇对细胞凋亡的影响,流式细胞术分析白藜芦醇对细胞周期分布和caspase-3活性的影响。结果低浓度(<30μmol/L)的白藜芦醇促进细胞增殖,而高浓度(>30μmol/L)抑制细胞增殖;白藜芦醇能够引起S/G2阻滞,并诱导细胞凋亡;白藜芦醇增加caspase-3活性具有时间和浓度效应。结论白藜芦醇促进或抑制细胞生长具有浓度依赖性;并通过增加caspase-3活性而诱导卵巢癌细胞凋亡。
Objective To investigate the effect of resveratrol on the growth and cell cycle of ovarian cancer cells, and to investigate the apoptosis of ovarian cancer cells induced by resveratrol. Methods MTT assay was used to detect the effect of resveratrol on cell growth. Ho33342 staining was used to observe the effect of resveratrol on nuclear condensation and fragmentation. TUNEL was used to detect the effect of resveratrol on cell apoptosis. The effect of resveratrol on cell cycle distribution and caspase-3 activity was analyzed. Results Resveratrol at low concentration (<30μmol/L) promoted cell proliferation, while high concentration (>30μmol/L) inhibited cell proliferation; resveratrol could induce S/G2 arrest and induced apoptosis; Resveratrol increases caspase-3 activity with time and concentration effects. Conclusion Resveratrol promotes or inhibits cell growth in a concentration-dependent manner, and induces apoptosis in ovarian cancer cells by increasing caspase-3 activity.
