目的掌握合肥市2012-2013年71例麻疹病例麻疹病毒流行特征及其基因分型。方法应用病原分子学逆转录-聚合酶链反应(RT-PCR),检测麻疹Ig M抗体阳性麻疹病例咽拭子样本。扩增麻疹病毒N基因COOH末端的634个核苷酸片段,扩增产物纯化后测序,基因分析采用MEGA 5.03软件。结果 2012、2013年麻疹病例麻疹病毒基因分析分别为8、63例,合计71例,RT-PCR扩增阳性麻疹毒株分别为2、23株,合计25株,研究分析均为H1a基因亚型。基因序列分析显示25株麻疹毒株分为2条传播主链,核苷酸平均变异为0%~2.4%(核苷酸差异0~11 bp)。25株麻疹毒株与Chin9322/H1a参考株核苷酸变异0.4%~1.6%(核苷酸差异2~6 bp),与S191疫苗株核苷酸变异7.6%~8.7%(核苷酸差异34~39 bp)。结论 H1a基因亚型是合肥市近年来麻疹传播流行优势毒株,毒株之间遗传变异程度小同源性高。
Objective To investigate the epidemiological characteristics and genotyping of measles virus in 71 measles cases from 2012 to 2013 in Hefei. Methods Throat swab samples of measles-positive IgM-positive measles patients were detected by reverse transcription-polymerase chain reaction (RT-PCR). The 634 nucleotides fragment of the COOH end of the measles virus N gene was amplified. The amplified product was purified and sequenced. The genetic analysis was performed using MEGA 5.03 software. Results The measles virus genetic analysis of measles cases in 2012 and 2013 were respectively 81 and 71. The total number of measles virus strains amplified by RT-PCR was 2,23 and the total number was 25, all of which were H1a genotypes . Sequence analysis showed that the 25 measles isolates were divided into two propagation backbones with average nucleotide variation of 0% -2.4% (nucleotide difference 0-11 bp). The nucleotide variation of 25 strains of measles and the strain of Chin9322 / H1a was 0.4% ~ 1.6% (nucleotide difference 2 ~ 6 bp), and that of the S191 vaccine strain was 7.6% ~ 8.7% (nucleotide difference 34 ~ 39 bp). Conclusion The subtype H1a is a epidemic predominant strain of measles transmission in Hefei in recent years. The degree of genetic variation among the strains is highly homologous.
