目的:观察IL-6mRNA及其蛋白在烧伤复合内毒素血症早期大鼠肝损害中的表达和细胞定位以及探讨IL-6在其发病过程中的作用。方法:选用156只Wistar大鼠随机分为复合组(C)、烧伤对照组(B)、内毒素对照组(E)和正常对照组(N)。应用血清定量、免疫组化(IHC)及原位杂交(ISH)等方法,检测了烧伤复合内毒素血症大鼠血清IL-6含量变化、肝组织IL-6和IL-6mRNA的细胞定位及其分布。结果:血清IL-6分别在0.5h、6~12h出现两个峰值(P<0.05),组织IL-6主要定位于肝窦内皮细胞(SECs)、枯否细胞(KCs),IL-6mRNA主要定位于KCs、中性粒细胞(PMNs)、巨噬细胞(MPs)。结论:提示IL-6是参与烧伤复合内毒素血症早期肝脏损害的重要细胞因子之一,并与肝细胞LBP的合成密切相关。
OBJECTIVE: To observe the expression of IL-6 mRNA and its protein in rat liver damage induced by burn complex endotoxemia and the role of IL-6 in its pathogenesis. Methods: 156 Wistar rats were randomly divided into compound group (C), burn control group (B), endotoxin control group (E) and normal control group (N). Serum levels of IL-6, IL-6 and IL-6 mRNA in liver tissues of burn rats were detected by serum quantification, immunohistochemistry (IHC) and in situ hybridization (ISH) Its distribution. Results: Serum IL-6 showed two peaks at 0.5h and 6-12h respectively (P <0.05). IL-6 mainly located in hepatic sinusoidal endothelial cells (SECs), Kupffer cells (KCs), IL -6 mRNA mainly located in KCs, neutrophils (PMNs), macrophages (MPs). Conclusion: It is suggested that IL-6 is one of the important cytokines involved in the early liver damage in burn endotoxemia and is closely related to the synthesis of LBP in hepatocytes.
