目的 检测黑色素瘤瘤小鼠高强度聚焦超声(HIFU)治疗前后局部(靶区及边缘)血管内皮生长因子(VEGF)、碱性成纤维细胞生长因子(bFGF)、转化生长因子(TGF) β1和TGF α等血管生长因子的表达,探讨HIFU治疗对局部肿瘤血管生长因子的影响。方法 采用B16黑色素瘤动物模型,实验分3组,即HIFU组、手术组和假照组。于治疗后第3天取各组靶区肿瘤组织(手术组除外)及边缘皮肤组织。采用免疫组织化学法检测VEGF、bFGF、TGF β1、TGF α的表达。结果 假照组靶区肿瘤组织VEGF、bFGF、TGF β1、TGF α表达阳性率分别为80 .0 %、5 0 .0 %、65 .0 %、80 .0 % ,而HIFU组为阴性表达;假照组边缘皮肤组织为阴性表达,而手术组及HIFU组边缘皮肤组织的上述4种血管生长因子的阳性表达率均为10 0 % ,两组差异无统计学意义(P >0 .0 5 )。结论 HIFU治疗能够破坏VEGF、bFGF、TGF β1、TGF α等血管生长因子在肿瘤组织中的表达,同时HIFU治疗也可以使靶区边缘血管生长因子的表达增加
Objective To detect the expression of vascular endothelial growth factor (VEGF), basic fibroblast growth factor (bFGF), transforming growth factor (TGFβ1) and fibroblast growth factor (bFGF) in melanoma tumor tissues before and after high intensity focused ultrasound (HIFU) TGF α and other vascular growth factor expression, explore the HIFU treatment of local tumor angiogenetic factors. Methods B16 melanoma animal model, the experiment divided into 3 groups, namely HIFU group, surgery group and sham group. On the 3rd day after treatment, the target tumor tissues (excluding the operation group) and the marginal skin tissues of each group were taken. Immunohistochemistry was used to detect the expression of VEGF, bFGF, TGFβ1 and TGFα. Results The positive rates of VEGF, bFGF, TGF β1 and TGF α in tumor tissue of the sham group were 80.0%, 50.0%, 65.0% and 80.0% respectively, but negative in HIFU group. The marginal skin tissues were negatively expressed in the sham group, while the positive expression rates of the above four vascular growth factors in the marginal skin tissues of the operation group and the HIFU group were both 100%, with no significant difference between the two groups (P> 0.05) ). Conclusions HIFU treatment can destroy the expression of VEGF, bFGF, TGFβ1, TGFα and other vascular growth factors in tumor tissues, and HIFU treatment can also increase the expression of vascular endothelial growth factor
