目的 研究酪氨酸激酶 (PTK)在类风湿关节炎 (rheumatoidarthritis,RA)成纤维样滑膜细胞 (fibroblast likesynoviocyte ,FLS)异常增生与转化特性中的作用。 方法 原代培养类风湿关节炎成纤维样滑膜细胞。应用形态观察、细胞计数、3H TdR掺入、流式细胞仪检测和双层琼脂培养法探讨染料木黄酮 ,特异性PTK抑制剂对于RAFLS细胞生长状态、数目、DNA合成、细胞周期及其非贴壁生长情况下细胞克隆形成数目的影响。结果 Genistein呈剂量依赖性抑制RAFLS的数目增长、3H TdR掺入和非贴壁生长的克隆形成数目 ;其生长抑制作用在细胞周期中G1期限制点。结论 酪氨酸激酶在RAFLS细胞异常增生与转化表现中发挥关键作用。
Objective To investigate the role of tyrosine kinase (PTK) in the abnormal proliferation and transformation of fibroblast like synoviocyte (FLS) in rheumatoid arthritis (RA). Methods Primary culture of rheumatoid arthritis fibroblast - like synoviocytes. Morphological observation, cell counting, 3H TdR incorporation, flow cytometry and double agar culture were used to investigate the effects of Genistein and specific PTK inhibitors on the growth status, number, DNA synthesis, cell cycle and cell cycle of RAFLS cells Effect of Wall Clone Formation Number on Wall Growth. Results Genistein inhibited the growth of RAFLS in a dose-dependent manner, the number of clones formed by 3H TdR incorporation and non-adherent growth, and the growth inhibition effect was at the G1 phase of the cell cycle. Conclusion Tyrosine kinase plays a key role in the abnormal proliferation and transformation of RAFLS cells.
