目的研制抗人滋养细胞单克隆抗体并分析其免疫生物学特性。方法采用滋养细胞免疫Balb/c小鼠 ;用细胞ELISA法鉴定mAb的特异性 ;用补体依赖的溶细胞作用测定mAb的细胞毒性。结果共获得8株分泌抗人滋养细胞mAb的杂交瘤细胞 ,其中2株mAb具有明显的细胞毒活性。抗滋养细胞mAb主要针对相对分子质量 (Mr)为80000和40000的两组滋养细胞表面抗原。免疫性流产患者的血清 (含抗滋养细胞抗体 )可阻断mAb与滋养细胞膜抗原的反应。结论利用细胞免疫法所获该组mAb所针对的抗原表位 ,可能是诱发免疫性流产的滋养细胞膜特异性抗原。
Objective To develop anti-human trophoblast monoclonal antibody and analyze its immunobiological properties. Methods Balb / c mice were immunized with trophoblast cells. The specificity of mAb was evaluated by ELISA. The cytotoxicity of mAb was determined by complement-dependent cytolysis. Results A total of 8 hybridoma cells secreting anti-human trophoblast mAb were obtained, of which 2 mAbs had significant cytotoxic activity. Anti-trophoblastic mAbs are directed against two groups of trophoblastic surface antigens with relative molecular mass (Mr) of 80,000 and 40,000. Serum (including anti-trophoblastic antibodies) in immunocompromised patients can block the mAb’s response to trophoblast antigens. CONCLUSION: The antigenic epitopes targeted by this group of mAbs obtained by cellular immunization may be trophoblast-specific antigens that induce immune abortion.
