目的 探讨子痫前期患者胎盘组织中线粒体解偶联蛋白2(UCP2)基因mRNA表达与胰岛素抵抗的关系.方法 检测36例正常足月妊娠妇女(A组)和24例轻度子痫前期患者(B组)及17例重度子痫前期患者(C组)剖宫产前空腹血及新生儿脐血中血糖、胰岛素水平,采用RT-PCR法测定胎盘组织中UCP2 mRNA表达水平.结果 C组新生儿脐血血糖和胰岛素显著低于A组(P<0.05);B、C组UCP2基因mRNA表达水平分别为(0.942±0.202)、(0.908±0.188),显著低于A组的(1. 908±0.283)(P<0.05).结论 子痫前期患者胎盘组织中UCP2的低表达在子痫前期的发病中有重要作用,可能参与了胰岛素抵抗的病理生理过程.“,”Objective To study the relationship between expression of uncoupling proteins 2 (UCP2) gene mRNA in the placenta and insulin resistance (IR) in preeclampsia patients. Methods Fasting maternal serum and umbilical cord blood insulin and blood glucose(Glu) were detected in 36 cases of normal pregnancy woman (group A) , 24 cases of mild preeclampsia (group B)and 17 cases of severe preeclampsia (group C). Expression of UCP2 gene mRNA in the placenta was detected by semi-quantitative RT-PCR. Results The levels of umbilical cord Glu and insulin were significantly lower in group C than those in group A (P<0. 05). Expressions of UCP2 gene mRNA in the placenta in groups of B and C were ( 0. 942 ± 0. 202 ) and ( 0. 908 ± 0. 188 ), respectively, which were significantly lower than (1. 908±0. 283) in group A (P<0. 05). Conclusion The low expression of UCP2 gene mRNA in the placenta of preeclampsia patients may induce IR, which may play an important role in the pathogenesis and development of preeclampsia.
