平皿培养用于供检验者观察菌落形态,当检出病原菌后就希望把这种培养物杀死。以前用来杀灭玻璃平皿培养物的氯仿能使塑料聚苯乙烯平皿发生变化,因而认为选用甲醛处理比较满意。要试验出杀灭平皿培养物的准确时间是困难的。1894年Klein最早在固体培养基中试验杀灭培养物。在检测大肠菌产物时已定出用氯仿杀灭培养物的时间范围为15分钟至2小时。Colline和Lyne在“这可能杀灭生物”的短评中推荐2小时。
The plate culture is used to visualize colony morphology for the test subject and it is desirable to kill the culture when the pathogen is detected.
