目的通过观察过氧化物酶体增殖物活化受体γ共激活因子-1α(PGC-1α)在中枢性肥胖小鼠中的表达水平研究PGC-1α与能量代谢和肥胖的相关性。方法皮下注射谷氨酸钠建立小鼠中枢性肥胖的病理模型,比较各组小鼠的BMI、摄食量及测定体温的变化;检测PGC-1α在棕与白色脂肪、肝脏、肺脏、心脏和脑中的表达水平。结果 PGC-1α在上述组织中都有不同程度的表达;而模型组PGC-1α的表达下调。PGC-1α在棕色脂肪的表达始终高于白色脂肪;寒冷刺激可诱导脂肪组织中PGC-1α表达增加,且在棕色脂肪中尤为显著。结论 PGC-1α可能与机体能量代谢和肥胖的发生密切相关。
Objective To investigate the correlation between PGC-1α and energy metabolism and obesity by observing the expression of peroxisome proliferator-activated receptor γ-coactivator-1α (PGC-1α) in central obese mice. Methods The mice model of central obesity was established by subcutaneous injection of sodium glutamate. The BMI, food intake and body temperature of mice in each group were compared. The changes of PGC-1α in brown and white fat, liver, lung, heart and brain In the expression level. Results The expression of PGC-1α in all the above tissues was different; however, the expression of PGC-1α in the model group was down-regulated. The expression of PGC-1α in brown fat was always higher than that of white fat; cold stimulation induced the increase of PGC-1α expression in adipose tissue, especially in brown fat. Conclusions PGC-1α may be closely related to the body’s energy metabolism and obesity.
