本文探讨体外培养的各种成纤维细胞在机械力的作用下,细胞骨架、生长方向、细胞增殖、细胞外基质和生长因子发生的变化,以及可能发生的信号传导途径变化。在机械应力作用下,成纤维细胞的形态和骨架发生了规律的改变;细胞的生长方向发生重排,且与力的方向呈垂直趋势;适宜范围内的机械力可以促进细胞的增殖;机械应力的刺激可引起细胞外基质合成增加和多种生长因子的分泌,导致组织适应性改变;而出现这些现象的机制,可能是机械力作用于细胞启动了MAPK、一氧化氮和氧化应激途径。
This article explores the changes of cytoskeleton, growth direction, cell proliferation, extracellular matrix and growth factors, and possible signal transduction pathways under the action of mechanical force of various fibroblasts cultured in vitro. Under the action of mechanical stress, the morphology and skeleton of fibroblasts changed regularly. The growth direction of cells was rearranged and showed a vertical trend with the direction of force. The suitable mechanical force could promote the proliferation of cells. The mechanical stress Stimulates the synthesis of extracellular matrix and the secretion of multiple growth factors, leading to the changes of tissue adaptability. The mechanism of these phenomena may be that the mechanical force acts on the cells to initiate the MAPK, nitric oxide and oxidative stress pathways.
